Identification of KO on DNA level. gDNA from KO and WT strains was used as the template. Primers for EHT1 gene, recombinant arm (left L/RF1 and right R/RF2), and selection marker (Hyg) amplified relevant sequences. In KO mutant, L/RF1, R/RF2, and Hyg should produce bands indicative of the insertion of the vector, while WT should lack vector sequence bands. EHT1 gene was not present in KO mutant compared with WT.
