PDK1 is a direct and functional target gene of miR-375. (a) miR-375 was sequenced to reflect the positioning of PDK1 3-UTR seed regions and associated mutations. (b) The dual-luciferase reporter assay was performed with the pmirGLO dual-luciferase vector in CNE-2 cells. Detection of relative levels of PDK1 mRNA (c) and protein (d) expression in CNE-1 and CNE-2 cells transfected with miR-375 mimic or inhibitor, as determined by qRT-PCR and Western blot. vs. miR-NC.
