Isolation and characterization of canine NK cells. (a) CD5 negative (CD5lo) cells were isolated from canine PBMCs and activated in specific culture supplemented with human IL-2 (500 U/mL), IL-15 (10 ng/mL), and canine IL-21 (5 ng/mL) for 21 days. (b) Cell surface markers were analyzed by FACS flow cytometric analysis at activated CD5lo cells. (c) To determine whether CD5lo cells develop into cytotoxic NK cells, the expression of activated NK cell markers (NKp30, NKp44, NKp46, NKG2D, CD244, Perforin 1, and Granzyme B) in activated canine CD5lo cells was analyzed by quantitative RT-PCR. β-Actin was used as a reference gene. , , .
