Inhibition of the mTORC1/NF-κB Axis Alters Amino Acid Metabolism in Human Hepatocytes

SC75741 inhibits amino acid catabolic gene expression in Rheb-overexpressing hepatocytes. Rheb-overexpressing hepatocytes were treated with 10 μM SC75741 for 12 h, and then, NF-κB phosphorylation and the expression levels of amino acid catabolic genes were determined. (a) NF-κB activation was enhanced by Rheb overexpression and inhibited by SC75741 treatment. (b) The expression levels of amino acid catabolic genes were upregulated by Rheb overexpression and inhibited by SC75741 treatment.
