Effects of DHTS on the induction of ER stress-related protein expression and XBP1 mRNA splicing in human prostate carcinoma cells. (a) DU145 cells were treated with 1.5 μg/mL DHTS for the indicated time periods, and (b) DU145 cells were treated with various concentrations of DHTS for 12 h (CHOP, p-eIF2α, p-JNK, and p-PERK) or 24 h (GRP78/Bip). Total cellular protein was subjected to Western blotting to detect the expressions of GRP78/Bip, CHOP/GADD153, phosphor-eIF2α, phosphor-JNK, and phosphor-PERK. (c) DU145 cells were treated with 1.5 μg/mL DHTS for the indicated time periods, and total RNA was collected for detecting XBP1 mRNA expression by RT-PCR.
