Effects of oleifoliosides A on LPS-stimulated phosphorylation and degradation of I B- and nuclear translocation of p65. (a) and (b) Cells were pretreated with the indicated concentrations of oleifoliosides A 1 h and then LPS (500 ng/mL) treatment for 15 or 60 min. Cytosol or nuclear protein was determined by Western blot assay using anti-p-I B- , I B- , NF- B p50, and NF- B p65. GAPDH and hnRNP were used as internal controls for Western blot. (c) The cells were pretreated with 15 μM oleifoliosides A for 1 h prior to stimulation with LPS (500 ng/mL) for 1 h. The nuclear localization of NF- B p65 was determined using confocal laser scanning microscopy after being stained with DAPI, anti-NF- B p65, and FITC-labeled anti-rabbit IgG antibody. per experiment.
