A Special Ingredient (VtR) Containing Oligostilbenes Isolated from Vitis thunbergii Prevents Bone Loss in Ovariectomized Mice: In Vitro and In Vivo Study

Figure 6

Effects of oligostilbenes isolated from Vitis thunbergii on the differentiation and function of primary cultured osteoblasts and osteoclasts, respectively. (a) (−)-Vitisin B and ampelopsin C concentration-dependently stimulated the increases in ALP activity and mineral matrix deposition (Alizarin Red staining) in osteoblasts, respectively. The cells were incubated with the tested drug for 7 or 14 days as described in Materials and Methods. Each value is the mean ± S.E.M. of six independent experiments, each in triplicate. * and ** , compared with the control group without the tested drug treatment. (b) (+)-Vitisin A concentration-dependently repressed RANKL-induced osteoclast differentiation, TRAP+MNCs formation, and bone resorption. Mouse BMM cells were cultured for 4 or 8 days in the presence of M-CSF and RANKL, as described in Materials and Methods.. Results are presented as means ± S.E.M. of four independent experiments. * , ** , and *** , as compared with RANKL.
