Naringenin Inhibits Adipogenesis and Reduces Insulin Sensitivity and Adiponectin Expression in Adipocytes

Naringenin modulates insulin action via attenuation of IRS-1 tyrosine896 phosphorylation. Mature 3T3-L1 adipocytes were serum deprived overnight and then pretreated with vehicle or 25 μg/mL naringenin (Nar) for 2 hours. The control (C) cells were untreated. As indicated, cells were then stimulated with 10 nM insulin for 2 minutes prior to preparation of whole cell extracts. One hundred fifty micrograms of protein from the whole cell extracts were separated by SDS-PAGE, transferred to nitrocellulose, and probed for IRS-1 tyrosine896 phosphorylation using a phosphospecific antibody directed against this site (a). Band intensities were quantified, normalized against the loading control, MAPK, and presented as a bar graph (b). This experiment was independently repeated three times.
