Ginsenoside RK3 Prevents Hypoxia-Reoxygenation Induced Apoptosis in H9c2 Cardiomyocytes via AKT and MAPK Pathway

Involvement of AKT signaling transduction pathway and RK3 in H/R-induced caspase-3 activation and cytotoxicity. H9c2 cardiomyocytes were pretreated with or without RK3 (25 μg/mL) for 12 h prior to H/R exposure. The expression levels of ERK1/2, p38, and JNK activity were detected by immunoblotting assay (a). The effects of LY294002 (an AKT-specific inhibitor) on RK3-mediated activation of HO-1, caspase-3, and cell viability in H9c2 cardiomyocytes were measured (b and c). The effect of Znpp (an inhibitor of HO-1 activity) on RK3-mediated cardioprotection was also evaluated by CCK8 assay (d).
