Sargassum fulvellum Protects HaCaT Cells and BALB/c Mice from UVB-Induced Proinflammatory Responses

Inhibition of UVB-induced COX-2 and TNF-αexpressions and subsequent PGE2 production by SFE-EtOAc in HaCaT cells. HaCaT cells were pretreated with vehicle or SFE-EtOAc (30 μg/mL and 100 μg/mL) for 9 h before irradiation of UVB (60 mJ/cm2). Twenty-four hours after UVB exposure (60 mJ/cm2), the protein expression of COX-2 and TNF-α (a) and accumulation of PGE2 into the media (b) were monitored by western blot analysis and ELISA, respectively. (a) Protein extracts were loaded onto a SDS-PAGE and then transferred to PVDF membranes. The membranes were probed with anti-COX-2, anti-TNF-α, or antiactin primary antibody. Actin levels were assessed to confirm the equal amount of protein loading. Representative blots from three independent experiments are shown. (b) The relative levels of the PGE2 were represented as fold induction in comparison with the vehicle-treated control cells. Asterisks indicate a significant difference from the group exposed to UVB alone (** ).
