Clitocybe nuda Activates Dendritic Cells and Acts as a DNA Vaccine Adjuvant

WE-CN-treated BMDCs increase T-cell activation in response to the specific antigen OVA. (a) CD8+ T cells and CD4+ T cells were prepared from the spleens of OT-I and OT-II mice, respectively. Purified T cells were cocultured with PBS-, LPS- (100 ng/mL), or WE-CN- (100 μg/mL) treated DCs pulsed with an OVA peptide at the indicated ratio of DC : T cells for 96 hr. Cell proliferation was quantified by [3H]-thymidine incorporation for 18 hr. (b) Supernatants were collected from cultures after 4 days, and IFN-γ production was measured by ELISA. The data shown represent the mean ± SD of samples from three wells. * compared to control. All data are representative of three independent experiments showing similar results.
