WE-CN induces MAPK phosphorylation and NF-κB binding activity in BMDCs. Immature BMDCs were treated with WE-CN (100 μg/mL), and whole cell lysates or the cytosolic fraction of lysates was collected at the indicated time points. (a) MAPK phosphorylation levels were analyzed by western blot analysis with anti-ERK, anti-JNK, and anti-p38 MAPK antibodies (against both phosphorylated and nonphosphorylated forms of the proteins). pErk, pp38, and pJNK 2 levels were normalized with total Erk, p38, and JNK levels and then compared with the control group arranged as one unit. The data represent the mean ± SD of samples from three independent experiments. (b) The NF-κB assay was described in Section 2. The NF-κB binding activity is shown as relative OD450 levels.
