CBF affects HIF-1α transcription and protein expression. (a) Elevation of HIF-1α transcription level by CBF in HCT116 cells. Compared with that in normal atmosphere, the level of HIF-1α mRNA doubled when HCT116 cells were in lack of oxygen. (b) Elevation of HIF-1α transcription level by CBF in HT29 cells. Dramatic upregulation of HIF-1α mRNA was only from 8 to 18 hour time points, followed by a sharp fall at 24 hours. The patterns of HIF-1α transcription of HT29 between 1% and 20% were similar. (c) Inhibition of HIF-1α protein expression. Under hypoxic (100 µM CoCl2) conditions, HIF-1α protein expression was suppressed by CBF at different time points. (d) Inhibition of GFP fused HIF-1α expression. Plasmid pGFP-HIF-1α was transfected into HCT116 and HT29 cells, followed by a 24-hour-CBF treatment under 1% or 20% oxygen. Compared with the control groups, 1 μM of CBF exposure effectively inhibited the expression of GFP under hypoxic conditions ( , * ).
