Icariin Prevents Amyloid Beta-Induced Apoptosis via the PI3K/Akt Pathway in PC-12 Cells

Figure 3

Involvement of PI3K/Akt signaling in the protective effects of icariin against Aβ25–35-induced cytotoxicity in PC12 cells. After 1 h pretreatment with the PI3K inhibitor LY294002 (50 μM), PC12 cells were treated with 20 μM icariin for 1 h, followed by Aβ25–35 treatment for 24 h. Cell viability and apoptosis were determined by MTT assay and Annexin V/PI double staining, respectively. (a) LY294002 inhibited the icariin-mediated increase in viability in Aβ25–35-treated cells. (b) LY294002 inhibited the icariin-mediated decrease in apoptosis in Aβ25–35-treated cells. (c) (A) Untreated control, (B) Aβ25–35 treatment, (C) icariin treatment, (D) LY294002 treatment, and (E) icariin + LY294002. Results are presented as mean ± S.D. of triplicate independent experiments. versus untreated control; versus Aβ25–35 treatment; versus icariin treatment.
