Involvement of p38 and PKC-δ signaling pathway in KRG-treated TrxR1 expression in HUVECs. Cells were pretreated with 20 μM SB203580 (p38 inhibitor) or 10 μM Rottlerin (PKC-δ inhibitor) for 1 h, followed by incubation with 1 mg/mL KRG for 18 h. Whole cell lysates were analyzed by Western blot with antibodies against TrxR1 and GAPDH (a and c). Transient transfection of cells with 10–30 nM p38 and PKC-δ siRNA suppressed the upregulation of TrxR1 expression by KRG (b and d).
