Maoto, a Traditional Japanese Herbal Medicine, Inhibits Uncoating of Influenza Virus

Maoto is not toxic for cell lines and influenza viruses. (a) A549 cells were cultured with 0 to 1,000 µg/ml maoto for 24 hr and then assayed by flow cytometry after cell staining with 7-amino-actinomycin D and Annexin V, from which the double negative population is determined as viable cells. (b) A dense concentration of PR8 (109 TCID50/ml) was incubated with medium only, or pretreated with 400 µg/ml maoto for 0.5 min or 60 min, and was used to infect A549 cells (moi = 1) followed by the culture for 24 hr. Culture supernatants were assayed for the titer of infectious virus.
