mRNA expression profile of Bax, p53, Bcl-2, and Bcl-XL in HL-60, MCF-7, and HT29 cell lines. A total of 1 × 10⁶ cells were treated with 10 μg/mL of alcoholic extracts for 24 h. Total RNA was isolated and treated with DNase; 1 μg of RNA was reverse-transcribed into cDNA with a synthesis kit, using oligo-dT and a random hexamer. mRNA levels were compared by RT-qPCR. Results were normalized to the β-globin gene and expressed as the mean ± SD relative to the negative control (C−, untreated cells). As positive control (C+) cells were treated with 7 μM of staurosporine. Experiments were done in triplicate. versus C−; versus C−.
