The object of this study is to observe the effects of platycodin D, a saponin purified from <i>Platycodi Radix</i>, on mice collagen-induced arthritis (CIA). A daily dose of 200, 100, and 50&#x2009;mg/kg platycodin D was administered orally to male DBA/1J mice for 40 days after initial collagen immunization. To ascertain the effects administering the collagen booster, CIA-related features (including body weight, poly-arthritis, knee and paw thickness, and paw weight increase) was measured from histopathological changes in the spleen, left popliteal lymph node, third digit, and the knee joint regions. CIA-related bone and cartilage damage improved significantly in the platycodin D-administered CIA mice. Additionally, myeloperoxidase (MPO) levels in the paw were reduced in platycodin D-treated CIA mice compared to CIA control groups. The level of malondialdehyde (MDA), an indicator of oxidative stress, decreased in a dose-dependent manner in the platycodin D group. Finally, the production of IL-6 and TNF-<i>&#x3b1;</i>, involved in rheumatoid arthritis pathogenesis, was suppressed by treatment with platycodin D. Taken together, these results suggest that platycodin D is a promising new effective antirheumatoid arthritis agent, exerting anti-inflammatory, antioxidative and immunomodulatory effects in CIA mice.

The Effects of Platycodin D, a Saponin Purified from <i>Platycodi Radix</i>, on Collagen-Induced DBA/1J Mouse Rheumatoid Arthritis

Myasthenia gravis (MG) is an autoimmune disease, of which the pathogenesis has remained unclear. At present, MG does not have any effective treatment with minor side effects. Jian Ji Ning (JJN), a traditional Chinese medicine formula consisting of 11 medicinal plants, has been used in the treatment of MG for many years. The present study aims to determine if the Chinese herbal medicine JJN could lighten the clinical symptoms of patients with MG via the regulation of differential microRNAs (miRNAs) expression in serum. JJN should be orally administered  twice a day for 6 months. In the efficacy evaluation adopting the Quantitative Myasthenia Gravis Score (QMG), we found that JJN could improve the clinical symptoms of patients with MG more effectively. Besides, we found that JJN could regulate differential miRNAs expression in serum of patients with MG. Accordingly, we speculate that the effects of JJN on improving clinical symptoms and blood test indicators of patients with MG may be due to its inhibition of apoptotic pathways of some immune cells and its connection with  the regulation of serum miRNAs of some patients. In conclusion, we believe that JJN has a reliable curative effect on patients with MG-induced neuropathologic changes.

Clinical Study of Effects of Jian Ji Ning, a Chinese Herbal Medicine Compound Preparation, in Treating Patients with Myasthenia Gravis via the Regulation of Differential MicroRNAs Expression in Serum

<i  >Aim.</i> We aimed to investigate and evaluate the preventive activity of puerarin on the ovalbumin-induced asthma rat model. <i  >Materials and Methods.</i> Male Wistar rats were sensitized intraperitoneally on days 0, 7, and 14 and challenged to ovalbumin intratracheally on day 21. Groups of sensitized rats were treated randomly either with placebo, puerarin, dexamethasone, or puerarin combined with dexamethasone, from days 15 to 20. Inflammatory markers, including cell counts in bronchoalveolar lavage fluid (BALF), inflammatory cytokines, histopathology, and coagulation parameters, such as coagulation tests and the activity of coagulation factors, were analyzed. <i  >Results.</i> Puerarin significantly inhibited the recruitment of inflammatory cells in BALF and lung tissue. At the same time, the release of IL-4, IL-10, and IFN-<i  >&#x3b3;</i> in serum and the expression of mRNAs in lung tissue homogenate were changed  by puerarin. Puerarin Attenuates Ovalbumin-Induced Lung Inflammation and Hemostatic Unbalance in Rat Asthma Model

Acupuncture has been used to treat various disorders in China and some other eastern countries for thousands of years. Nowadays, acupuncture is gradually accepted as an alternative and complementary method in western countries for its undeniable therapeutic effects. However, its central mechanism is still unclear. It is especially difficult to reveal how different regions in the brain influence one another and how the relationship is among these regions responding to acupuncture treatment. Recently, by applying neuroimaging techniques and network theory, acupuncture studies can make further efforts to investigate the influence of acupuncture on regional cerebral functional connectivity (FC) and the modulation on &#x201c;acupuncture-related&#x201d; networks. Connectomics appears to be a new direction in research to further understand the central mechanism underlying acupuncture. In this paper, an overview of connectomics application in acupuncture research will be discussed, with special emphasis on present findings of acupuncture and its influence on cerebral FC. Firstly, the connectomics concept and its significance on acupuncture will be outlined. Secondly, the commonly used brain imaging techniques will be briefly introduced. Thirdly, the influence of acupuncture on FC will be discussed in greater detail. Finally, the possible direction in forthcoming research will be reviewed by analyzing the limitation of present studies.

Connectomics: A New Direction in Research to Understand the Mechanism of Acupuncture

Growing rates of fungal infections and increasing resistance against standard antifungal drugs can cause serious health problems. There is, therefore, increasing interest in the potential use of medicinal plants as novel antifungal agents. This study investigates the antifungal properties of crude plant extracts from ten medicinal plant species. Crude samples were extracted using the hot water extraction process. The minimum inhibitory concentrations (MIC) and diameter zone of inhibition were determined in each extract against ten fungal strains, and fluconazole was used as a positive control. The cytotoxicity of crude extracts on <i>in vitro</i> human skin fibroblast (HSF) cell models was determined by MTT assay. Of the ten crude extracts, <i>Psidium guajava</i> L. exhibited the highest antifungal activity, diameter zone of inhibition, and percentage HSF cell viability. Although all extracts exhibited antifungal activity, <i>Psidium guajava L</i>. had the greatest potential for developing antifungal treatments.
