Bufalin Inhibits HCT116 Colon Cancer Cells and Its Orthotopic Xenograft Tumor in Mice Model through Genes Related to Apoptotic and PTEN/AKT Pathways

Expression of PTEN/p-PTEN, AKT/p-AKT, Bad, Caspase-3/Caspase-3 cleaved in control, and Bufalin-treated cells. The expression of PTEN was upregulated in cells treated with 3 μM Bufalin, while the p-PTEN were significantly decreased in cells treated with 0.3 μM and 3 μM Bufalin compared to the control group (). There was significant downregulation of p-AKT in cells treated with 0.3 μM and 3 μM Bufalin (). The Bad expression was increased () in Bufalin-treated cells. The treatment of 0.3 μM and 3 μM Bufalin caused cleaves of Caspase-3 (fragments with molecular weight of 17 or 19 kd).
