Results of flow cytometry assay. HeLa cells were cotransfected by pEGFP-Wt-PSEN1(400) or pEGFP-Mut-PSEN1(400), pDsRed-N1, and indicated siRNAs, using Lipofectamine 2000 as a transfection reagent (details described in Section 2). HeLa cells containing both, the EGFP and RFP fluorescence, were counted in the BD FACS Calibur Flow Cytometry System (Becton Dickinson) using Ar-ion laser (488 nm). Fluorescence dot plots were generated using Cell Quest software.
