Sunitinib Combined with Th1 Cytokines Potentiates Apoptosis in Human Breast Cancer Cells and Suppresses Tumor Growth in a Murine Model of HER-2pos Breast Cancer

Metabolic activity of breast cancer cell lines is maximally suppressed by Th1 cytokines and sunitinib treatment. MDA-MB-468, MDA-MB-231, SKBR-3, HCC-1419, and MCF-7 cells were seeded at cells per well and were left either untreated (No Rx) or treated with Th1 cytokines (5 ng/mL each IFN-γ and TNF-α), sunitinib (10 μM), or sunitinib and Th1 cytokines (both). After 72 hours of incubation, Alamar Blue dye was added to each well. When a color change became apparent, the optical density of the supernatants was read via spectrophotometry at 630 nm. Data represent the summary analysis of at least three separate experiments per cell line. Statistical significance was determined by one-way ANOVA: MDA-MB-231 (, ), MDA-MB-468 (, ), MCF-7 (, ), HCC-1419 (, ), and SKBR-3 (, ), followed by the post hoc Tukey.
