Role of BMP-4 and Its Signaling Pathways in Cultured Human Melanocytes

BMP-4 activates Smad 1/5/8 and MAPK/ERK in cultured melanocytes. (a) Paired cultures of melanocytes were treated with vehicle or BMP-4 (25 ng/mL) for 2 and 4 hours. At each time point, cells were harvested and immunoblot analysis using polyclonal antibody against phosphorylated Smad 1/5/8 was performed. As the loading control, the membrane was immunoblotted for actin. One representative result from three separate experiments is shown. (b) Paired cultures of melanocytes were treated with vehicle or BMP-4 (25 ng/mL) for 15 and 30 minutes. At each time point, cells were harvested and immunoblot analysis using polyclonal antibody against phosphorylated and nonphosphorylated ERK was performed; PDBu at 1 × 1 0 6  M was used as a control. As the loading control, the membrane was immunoblotted for actin. One representative result from three separate experiments is shown.
