In order to determine whether BMP-4 affects promoter activity of PKC-, paired cultures of LH melanoma cells were transfected with PKC- promoter CAT reporter construct along with promoter-less renilla luciferase to assess the transfection efficiency, and treated with BMP-4 for 48 hours. Cells were harvested and CAT and Luciferase activities were performed. A total of eight separate transfections were performed, data were pulled and expressed as a percent of vehicle-treated samples. BMP-4 decreased the promoter activity of PKC- by 25 ±5% ().
