Role of BMP-4 and Its Signaling Pathways in Cultured Human Melanocytes

Effects of BMP-4 on the promoter activity of 𝑃 𝐾 𝐶 - 𝛽 . In order to determine whether BMP-4 affects promoter activity of PKC- 𝛽 , paired cultures of LH melanoma cells were transfected with PKC- 𝛽 promoter CAT reporter construct along with promoter-less renilla luciferase to assess the transfection efficiency, and treated with BMP-4 for 48 hours. Cells were harvested and CAT and Luciferase activities were performed. A total of eight separate transfections were performed, data were pulled and expressed as a percent of vehicle-treated samples. BMP-4 decreased the promoter activity of PKC- 𝛽 by 25 ±5% ( 𝑃 < . 0 1 ).
