CKIγ2 overexpression in fibroblasts reduces cell proliferation. (a) Cell proliferation was examined by seeding each cell line in triplicate at 3 × 105 cells/60 mm plate on day 0. On days 3, 5, and 7, cells were trypsinized and counted. Results are expressed as the average ± SEM of three independent experiments and for Z6 and Z23, the symbols include the SEM. *Significantly different at least at as determined by Student’s -test when compared with R2Zeo cells. (b) Incorporation of 3H-thymidine in response to PDGF (25 ng/mL) or serum (2.5% FBS) stimulation was performed in indicated cell lines. Results are expressed as the fold of thymidine incorporation in presence of PDGF or FBS over basal unstimulated condition and are the mean ± SEM of three independent experiments performed in quadruplicate. *Significantly different at least at as determined by Student’s -test compared with respective basal condition. (c) Indicated serum growing cells were submitted to DNA laddering assays according to standard procedures described in Materials and Methods. As positive control for apoptosis, primary rat thymocytes in culture were exposed 24 hours to anisomycin (10 μg/mL).
