Methods for Imaging Renin-Synthesizing, -Storing, and -Secreting Cells

Figure 3

Light micrographs of a cryostat kidney section. Renal tissue was obtained from a transgenic heterozygous (Ren -GFP/+) adult mouse in which green fluorescent protein (GFP) was placed under the control of the Ren renin locus by homologous recombination [25, 72]. Renin synthesis was stimulated by a 7-day-treatment with the angiotensin II type-1 receptor antagonist, losartan (25 mg/kg per day). (a) brightfield view showing the outline of an afferent arteriole (AA) and a glomerulus (GL) among proximal tubules (PT). (b) fluorescence view of the same field (white outline) showing green GFP signal from renin-synthesizing cells at the glomerular vascular pole (single arrow), and from upstream recruited cells (dashed arrows). Bar: 100  m.
