Effects of salubrinal on the phosphorylation of eIF-2α in RAW264.7 cells. (a) The cells were treated with 0 (DMSO alone), 5, and 10 μM salubrinal for 6 h. Phosphorylation of eIF-2α was analyzed by Western blotting. (b) The cells were stimulated with 10 μM salubrinal or DMSO alone for 6 h. Total RNA extracted from macrophages was converted to cDNA to determine the levels of TNF-α mRNA by PCR. The values are corrected with the levels of GAPDH mRNA. Values: means ± SEM ( ). (c) The cells were stimulated with 100 ng/mL LPS in the presence of 10 μM salubrinal or DMSO alone for 6 h. Phosphorylation of eIF-2α was analyzed by Western blotting.
