IFN-γ Elispot response to mitogens in lean, overweight, and obese chimpanzees. PBMCs isolated from the blood samples of the chimpanzees. Triplicate wells of the 96-well microtiter plates, precoated with IFN-γ antibody, were seeded with 105 PBMCs from the monkeys and incubated with 5 μg of each of the mitogens for 36 h at 37°C. At the end of the incubation period, the wells were washed and stained with biotinylated second IFN-γ antibody. The total number of spot forming cells in each of the mitogen-stimulated wells was counted and adjusted to control medium as background. See method section for experiment details. The results shown are an average of two distinct experiments and comparison between groups of chimpanzees was done by one-way analysis of variance with the Kruskal-Wallis test as described in Figure 1.
