Photochemical analysis of purified Arabidopsis phyB proteins assembled with phycocyanobilin (PCB). (a) Purification of PCB-assembled phyB proteins by streptavidin affinity chromatography. SDS-PAGE gel was shown with Coomassie blue staining. Lane S: Fermentas prestained protein ladder; lane C: crude extract; lane F: flow-through from the affinity column; lanes 2–5: elution fractions of holo-phyB. Zinc-induced fluorescence assay (Zinc) was shown to confirm chromophore-assembled phyB proteins. (b) Absorption and difference spectra of the purified PCB-assembled phyB proteins.
