Amine Analysis Using AlexaFluor 488 Succinimidyl Ester and Capillary Electrophoresis with Laser-Induced Fluorescence

Effect of varying DIEA concentration on peak intensity (a measure of labeling efficiency). Relative fluorescence intensities of dodecylamine are plotted against varying DIEA concentration. Labeling was conducted at 1 μM amine, 25 μM AF488 with the indicated concentration of DIEA in ethanol and incubated overnight. For analysis, the labeling solutions were diluted 1 : 10 in separation buffer (10 mM carbonate, 12 mM SDS, pH 10). Error bars are calculated from triplicate labeling at 12.5 μM.
