Total intracellular cAMP content in rat mesangial cells after chronic exposure (8 weeks) to high glucose with and without treatment with glibenclamide. Determinations were done under basal conditions and after 5 minutes treatment with 10 μM of the adenylyl cyclase activator forskolin, with or without a 15-minute preincubation with 0.5 mM phosphodiesterase inhibitor 3-isobutyl-1-methylxanthine (IBMX) to evaluate the role of PDE activity on cAMP accumulation. per experimental group.
