Application of IgG-Derived Natural Treg Epitopes (IgG Tregitopes) to Antigen-Specific Tolerance Induction in a Murine Model of Type 1 Diabetes

Treatment with Tregitopes in IFA at onset of diabetes. Female NOD mice were monitored for blood glucose (BG) levels. Disease onset was defined as BG between 200 and 250 mg/dL measured on two consecutive days, at which time mice were entered into the study. Mice received a single subcutaneous injection in the flank of either mTregitopes 167 and 289 (total 20 μg) formulated in IFA or PBS in IFA (as a vehicle control) or they were left untreated. BG levels were then measured weekly. In the initial study (a), Tregitopes suppressed the progression of diabetes in seven of twelve mice, whereas IFA alone had less of an effect. This study was repeated (b), showing successful “cure” of diabetes after onset. All pair-wise comparisons are significant ( ).
