SA inhibited the expression of Sirt1 and increased acetylated FoxO1. (a) Hepatocytes isolated from C57BL/6 mice were incubated with 5 or 10 μM SA for 24 h, and the expression of Sirt1 mRNA was analyzed by RT-qPCR, with values normalized to cyclophilin expression. The fold change was calculated as ratio of the expression in untreated hepatocytes. (b) Hepatocytes were cultured for 48 h with 5 or 10 μM SA. Western blot was performed with antibodies against Sirt1, FoxO1, and acetylated FoxO1 proteins. Data are mean ± SE from three independent experiments. compared with untreated hepatocytes.
