Aminoacylation assays. (a) Measurements of the aminoacylation levels in vivo. Two μg of total mitochondrial RNA purified from various cell lines under acid conditions was electrophoresed, electroblotted, and hybridized with a DIG-labeled oligonucleotide probe specific for the tRNAThr. The blots were then stripped and rehybridized with probes specific for the tRNAIle and tRNAHis, respectively. Average relative tRNAThr aminoacylation (Thr-tRNAThr) content per cell was normalized to the average content per cell of total tRNAThr (Thr-tRNAThr+tRNAThr) in mutant and control cybrid cell lines. (b) The proportion of aminoacylated tRNAThr in the mutant and control cybrid cell lines. The calculations were based on three independent determinations.
