fMLP-Induced IL-8 Release Is Dependent on NADPH Oxidase in Human Neutrophils

Amiloride reduces IL-8 and ROS production induced by fMLP. Neutrophils were incubated with vehicle or amiloride (100 or 500 μM) for 30 min and stimulated with fMLP for 4 h. IL-8 production was analysed in the supernatants by ELISA (a). Neutrophils were incubated with vehicle or amiloride (100 or 500 μM) for 10 min and then stimulated with 100 nM fMLP and the ROS production was detected by luminescence assay (b).
