Expression of ACAP1 by human PMNs. Lysates from RBL-2H3 cells (0.5 × 10⁶) overexpressing ACAP1-GFP and human PMNs (2 × 10⁶) were subjected to 8% SDS-PAGE and proteins were transferred to Immobilon PVDF membrane. Membranes were incubated with our homemade polyclonal antibodies (serums 161 and 162) against ACAP1 (1 : 1000) and exposed to peroxidase-conjugated anti-rabbit IgG (1 : 20,000) for 1 h at 37°C. The membranes were covered with ECL+ detection reagents. Images were obtained by exposing Kodak X-Omat film to membranes.
