Functional analysis of increased CysLT1 expression on T cells. The biological significance of the enhanced CysLT1 receptor expression on T cells was assessed by measuring intracellular calcium flux in response to LTD4. PBMC from HDM-allergic (HDMA) individuals were exposed for 72 h to Der p in the presence or absence of neutralizing Abs to IL-4 or IL-10 or of the CysLT1 receptor antagonist MK571. Cells were then labeled with anti-CD3 and anti-CD4 Ab and loaded with Fluo4-AM for the calcium mobilization assay on a FACSCalibur flow cytometer.
