Effect of TNF-α mAb on pro- and anti-inflammatory cytokine mRNA expression in LPS-stimulated monocytes. Each figure illustrates a time course experiment showing cytokine mRNA expression by monocytes stimulated with LPS (100 ng/mL) ± TNF-α mAb (10 μg/mL). Data points are expressed as median (range). AUC values were compared using the Wilcoxon Signed-Rank Test. values are one-tailed and are shown if significant. A significant reduction in AUC in the presence of TNF-α mAb was observed for CXCL8, IL-1β, IL-6, and IL-10 (). The experiments represent data from 4 subjects. Time points were conducted singly in culture wells and the resultant cellular material in duplicate for subsequent real-time PCR.
