(a) T-cell clone specificity was confirmed by incubating the T-cell clones with HLA-DR1+/HLA-A2− moDCs that were either pulsed or not pulsed with HLA-A2 derived peptides. (b) DR1+/A2− moDCs were incubated with different concentrations of monomeric HLA-A2 (A2) or the CD206-A HLA-A2 immune-complex (CD206-A-A2 ratio 3 : 1). T-cells were added to the culture following 4 h incubation with the antigen and subsequent washing steps. IFN-γ was measured in the supernatants after 24 h. Each experiment was conducted at least 3 times; shown is the mean of one experiment (performed in triplicate) with SD. ND: nondetectable.
