Quantification of the ADCC activity of TNF antagonists using iLite effector cells and mTNFα⁺ or mTNFα⁻ WT HEK293 target cell. iLite effector cells (1.2 × 10⁵ cells/well) were incubated with 2 × 10⁴ mTNFα target cells for 6 hours in the presence of increasing concentrations of infliximab, adalimumab, or etanercept prior to the addition of Nano-Glo Dual-luciferase reagent (Promega, Madison, WI) and the sequential determination of FL and NL activity. Results are expressed as FL activity in RLU.
