Ethyl Caffeate Ameliorates Collagen-Induced Arthritis by Suppressing Th1 Immune Response

ECF treatment markedly suppresses Th1 cell response in CIA. Splenocytes from CIA mice were restimulated for 4 h with PMA and ionomycin in the presence of Brefeldin A. At the end of incubation, cells were collected and blocked with rat-anti-mouse CD16/CD32, and then those were stained with PE-Cy7-anti-mCD4 antibodies; cells were fixed, permeabilized, and stained intracellularly with fluorochrome-conjugated anti-mIL-17A, anti-mIFN-γ. Samples were acquired on flow cytometer and analyzed by FlowJo software. Results presented are mean ± s.e.m., . , versus vehicle group.
