The Immunotherapeutic Effect of SIRPα-Silenced DCs against Cervical Cancer

Cytotoxicity assay. The DC-induced CTL response against the HPV16E7-expressing tumor was determined using a CCK8 kit. After 2 weeks of immunization, the pooled splenocytes from each group were used as effector cells. Their specific lytic activities against TC-1 cells at 90 : 1, 30 : 1, and 10 : 1 ratios were assayed. Ad-shSIRPα-transduced DCs induced the strongest specific CTL responses against TC-1 cells compared with the Ad-shControl group. The lytic activities against TC-1 of Ad-shSIRPα-transduced DCs were similar to those of the Ad-shControl group after CD8+T cell elimination with anti-CD8 monoclonal antibody.
