Anti-Inflammatory Activity of Diterpenoids from Celastrus orbiculatus in Lipopolysaccharide-Stimulated RAW264.7 Cells
Figure 6
Compounds 1 and 3 downregulated proinflammatory mediators. (a–c) The mRNA expression levels of IL-1β, IL-6, and TNF-α were measured using quantitative real-time PCR experiment, and these proinflammatory cytokines were significantly diminished by compounds 1 and 3. Cells were preincubated for 2 h with compounds 1 and 3 at concentration of 5 and 10 μM, respectively, and activated by LPS (1 μg/mL) for 2 h. Results represent as , and dexamethasone was used as a positive control. #, ###, , , and indicated significant differences from the LPS-treated group. (d) Graphical depiction of the potent anti-inflammatory activity of compounds 1 and 3 in LPS-activated RAW264.7 cells by suppressing the MAPK and NF-κB signaling pathway.
