Helicobacter pylori Infection Elicits Type I Interferon Response in Human Monocytes via Toll-Like Receptor 8 Signaling

Effect of TLR7/8 antagonists on TLR7 and TLR8 expression THP-1 cells. THP-1 cells were uninfected (mock), infected with H. pylori J99 strain at MOI 10 for 16 hours (J99), or treated with TLR7 inhibitor (ODN20958 + J99) or TLR7/8 inhibitor (ODN20959 + J99) for 4 hours prior to infection. Expression levels of TLR7 transcript were downregulated by both ODN20958 and ODN20959, whereas the expression of TLR8 transcript was downregulated by ODN20959 alone, suggesting specificity of the antagonists. Data is presented as from one experiment run in triplicate and is representative of two independent experiments. Statistical significance was analyzed using Student’s -test (; ; ; ns: not significant).
