Characterization of DOTAP liposomes and lipoplexes. The DOTAP liposomes were prepared using the direct-mixing and freeze-thaw-extrusion methods. Sizes of DOTAP liposomes or lipoplexes prepared by (a) the direct-mixing method and by (b-c) the freeze-thaw-extrusion method with (b) 100 nm or (c) 1000 nm filter. Data were shown as means ± s.e. (d) DNA complexation efficiency. The DNA : DOTAP ratio was 1 : 12 (w/w). The time indicates the duration of complexation with DNA.
