A Ploidy Increase Promotes Sensitivity of Glioma Stem Cells to Aurora Kinases Inhibition

Danusertib induces a reduction of phosphorylated Aurora kinases in all the GSCs. Representative images (up) of untreated or 48 h 500 nM Danusertib treated sensitive (GBM2) and resistant (G166) GSC lines, synchronized with STLC and stained for Crest (white), phospho-Aurora kinases (red), γ tubulin (green), and DAPI (blue) are reported. Scale bar = 5 μm. Quantification of the pospho-Aurora kinases fluorescent signal in the nuclei of mitotic cells was performed by ImageJ software (bottom). Results are expressed as mean of two independent experiments. At least 25 cells were analyzed in each experiment. Error bars represent SEM. t- test was performed on raw data: p<0,001.
