Inhibition of Proteasome Activity Upregulates IL-6 Expression in RPE Cells through the Activation of P38 MAPKs

Protease inhibitor MG132 upregulated the protein level of IL-6 in the medium of RPE in MG132 group compared with that in the control group, but it reduced the secretion of MCP-1. (a) Levels of IL-6 in the medium were detected by ELISA following incubation of ARPE-19 cells in the presence or absence of MG132 (10 μM) for 2, 4, 6, 8, 10, or 12 h. (b) Levels of IL-6 in the medium were detected by ELISA during 0 h–6 h or 6 h–12 h in the presence or absence of MG132 (10 μM). (c) Levels of MCP-1 in the medium were detected by ELISA following incubation of ARPE-19 cells in the presence or absence of MG132 (10 μM) for 2, 4, 6, 8, 10, or 12 h. (d) Levels of MCP-1 in the medium were detected by ELISA during 0 h–6 h or 6 h–12 h in the presence or absence of MG132 (10 μM). The results are the mean ± SD and as compared with the control.
