The complex <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mrow><mml:mo>[</mml:mo><mml:mrow><mml:msub><mml:mrow><mml:mtext>Pt</mml:mtext></mml:mrow><mml:mn>2</mml:mn></mml:msub><mml:msub><mml:mrow><mml:mtext>Cl</mml:mtext></mml:mrow><mml:mn>2</mml:mn></mml:msub><mml:msub><mml:mrow><mml:mrow><mml:mo>(</mml:mo><mml:mrow><mml:mtext>Spym</mml:mtext></mml:mrow><mml:mo>)</mml:mo></mml:mrow></mml:mrow><mml:mn>4</mml:mn></mml:msub></mml:mrow><mml:mo>]</mml:mo></mml:mrow></mml:math>, where Spym &#61; 2-mercaptopyrimidine, was synthesized andanalyzed spectroscopically. The presence in the <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mmultiscripts><mml:mtext>P</mml:mtext><mml:mprescripts /><mml:none /><mml:mrow><mml:mn>195</mml:mn></mml:mrow></mml:mmultiscripts><mml:mtext>t</mml:mtext></mml:math> NMR spectrum, of only one signal for the Pt(III) indicates the symmetrical arrangement of the ligands and the identical setting of N, S and Cl atoms, <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>PtS</mml:mtext><mml:mtext>2</mml:mtext></mml:msub><mml:msub><mml:mtext>ClN</mml:mtext><mml:mn>2</mml:mn></mml:msub></mml:math>, for the two Pt atoms being different to other compounds described in theliterature. The interaction of this complex with DNA was studied by several techniques, including circular dichroism, melting temperature determination, electron microscopy (EM) and atomic forcemicroscopy (TMAFM). Preliminary results show a high activity against HL-60 and HeLa tumour lines for the Pt-2-mercaptopyrimidine complex in comparison with cisplatin activity. Higher values for <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>IC</mml:mtext><mml:mrow><mml:mn>50</mml:mn></mml:mrow></mml:msub></mml:math> were obtained, while the values of <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>LD</mml:mtext><mml:mrow><mml:mn>50</mml:mn></mml:mrow></mml:msub></mml:math> were lower than those for cisplatin.

Antitumour Activity of a <small.letters>pt</small.letters>(III) Derivative of 2-Mercaptopyrimidine

Cisplatin is an extremely effective cancer chemotherapeutic agent, but its use is often accompanied by toxicity. Second generation drugs such as carboplatin are becoming more widely used because of reduced toxicity. Since biotransformation products have been implicated in the toxic responses, we have begun to investigate the reactions of cisplatin and carboplatin with potential biological ligands. Human urine samples from patients on cisplatin chemotherapy were shown to contain cisplatin, its hydrolysis product and biotransformation products containing Pt-creatinine, Pt-urea and Pt-uric acid complexes. Urine from carboplatin patients shows fewer biotransformation products. Studies with control and diabetic (protected against cisplatin toxicity) rats showed systematic differences in the biotransformation products formed on administration of cisplatin.

Determination of Biotransformation Products of Platinum Drugs in Rat and Human Urine

Nickel(II) complexes of thiosemicarbazons were observed to be potent cytotoxic agents in human and rodent tissue cultured tumor cells. Each compound demonstrated a slightly different profile in the various histological types of tumors. The nickel complex of Appip demonstrated the most potent in vivo activity in the Ehrlich ascites carcinoma. This agent selectively inhibited L1210 DNA and purine syntheses, and DNA polymerase &#945;, PRPP-amido transferase, IMP-dehydrogenase, dihydrofolate reductase, TMP-kinase and thymidylate synthetase activities. L1210 DNA strand scission was evident and DNA viscosity was reduced after 24 hr incubation. The nickel complexes were not L1210 DNA topoisomerase II inhibitors.

The in vitro anti-tumour activity of 1, determined against a variety of cell lines, is compared with those of the corresponding 2-methylcarboranylacetate, derivative 2, and with clinically used compounds.

X-Ray Structure and In Vitro Anti-Tumoural Activity of the Dimeric Bis&#91;(2-Phenyl-1,2-Dicarba-Closo-Dodecaborane-1-Carboxylato)-Di-<small.letters>n</small.letters>-Butyltin&#93; Oxide

Several cobalt(II), copper(II) and nickel(II) complexes of nicotinoylhydrazine-derived compounds were prepared and characterised by physical, spectral and analytical data. These compounds and their complexes have proven to be antibacterial. The screening data show the metal complexes to be more potential/bactericidal than the uncomplexed compounds against one or more bacterial species.

The ternary complexes of formulae cis-&#91;<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mrow><mml:mo>(</mml:mo><mml:mrow><mml:msub><mml:mrow><mml:mtext>NH</mml:mtext></mml:mrow><mml:mn>3</mml:mn></mml:msub></mml:mrow><mml:mo>)</mml:mo></mml:mrow><mml:mn>2</mml:mn></mml:msub></mml:math>Pt(nucl)(amac)&#93;<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>NO</mml:mtext><mml:mn>3</mml:mn></mml:msub></mml:math>, where nucl &#61; guo and cyd (guanosine and cytidine) and amac &#61; the deprotonated aminoacids glycine (gly), L-alanine (ala), L-2-aminobutyric acid (2-aba), L-norvaline (nval) and L-norleucine (nleu), were prepared from the reactions of the binary chelated ones cis-&#91;<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mrow><mml:mo>(</mml:mo><mml:mrow><mml:msub><mml:mrow><mml:mtext>NH</mml:mtext></mml:mrow><mml:mn>3</mml:mn></mml:msub></mml:mrow><mml:mo>)</mml:mo></mml:mrow><mml:mn>2</mml:mn></mml:msub></mml:math>Pt(amac)&#93;<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>NO</mml:mtext><mml:mn>3</mml:mn></mml:msub></mml:math> with the nucleosides.They were characterized by <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mmultiscripts><mml:mtext>H</mml:mtext><mml:mprescripts /><mml:none /><mml:mn>1</mml:mn></mml:mmultiscripts></mml:math>, <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mmultiscripts><mml:mtext>C</mml:mtext><mml:mprescripts /><mml:none /><mml:mrow><mml:mn>13</mml:mn></mml:mrow></mml:mmultiscripts></mml:math> and <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mmultiscripts><mml:mtext>Pt</mml:mtext><mml:mprescripts /><mml:none /><mml:mn>195</mml:mn></mml:mmultiscripts></mml:math> NMR and IR spectra, together with elemental analysis and conductivity measurements. The aminoacids coordinate with Pt(II) in the ternary complexes with their terminal -<mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>NH</mml:mtext><mml:mn>2</mml:mn></mml:msub></mml:math> groups, guo through <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>N</mml:mtext><mml:mn>7</mml:mn></mml:msub></mml:math> and cyd through <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>N</mml:mtext><mml:mn>3</mml:mn></mml:msub></mml:math>. Ligand-ligand hydrophobic interactions were also observed in the ternary complexes and were stronger with longer aliphatic chains of the aminoacids. The <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:mmultiscripts><mml:mi>E</mml:mi><mml:mprescripts /><mml:none /><mml:mn>3</mml:mn></mml:mmultiscripts></mml:math> sugar conformation increased by 5-7&#37; in the ternary systems, as compared to the free nucleosides, while the percentage of the gg conformation remained almost constant and the one of the anti conformation of the sugar increased also slightly.Finally, the h conformer around the <mml:math xmlns:mml="http://www.w3.org/1998/Math/MathML"><mml:msub><mml:mtext>C</mml:mtext><mml:mtext>&#x03B1;</mml:mtext></mml:msub><mml:msub><mml:mtext>-C</mml:mtext><mml:mtext>&#x03B2;</mml:mtext></mml:msub></mml:math> bonds of the aminoacids reached a maximum in the binary systems and decreased again considerably in the ternary ones.

Complexes containing the anions of 5-benzoylamido-1,3,4-thiadiazole-2-sulfonamide and 5-(3-nitro-benzoylamido)-1,3,4-thiadiazole-2-sulfonamid as ligands, and V(IV); Cr(III); Fe(III); Co(II); Ni(II); Cu(II) and Ag(I) were synthesized and characterized by standard procedures (elemental analysis; IR, electronic, and EPR spectroscopy; TG, magnetic and conductimetric measurements). The original sulfonamides and their metal complexes are strong inhibitors of two carbonic anhydrase (CA) isozymes, CA I and II.
