Cathepsin G, a Neutrophil Protease, Induces Compact Cell-Cell Adhesion in MCF-7 Human Breast Cancer Cells

Cathepsin G induces cell condensation in MCF-7 cells on fibronectin, but not on type IV collagen. MCF-7 cells were cultured in dishes coated with fibronectin or type IV collagen for 24 hours. After washing, adherent cells were incubated in serum-free medium without or with 0.5 mU/mL cathepsin G. (a) Cathepsin G-induced cell condensation of MCF-7 cells was analyzed by phase-contrast microscopy in the indicated periods as described in Materials and Methods. (b) MTT-reducing activity was measured at the indicated times. Bars represent the standard deviation. Analyses were performed in triplicate in duplicate experiments.
