Inhibition of Toll-Like Receptor 2-Mediated Interleukin-8 Production in Cystic Fibrosis Airway Epithelial Cells via the 𝛼7-Nicotinic Acetylcholine Receptor

Inhibition of α7-nAChR abrogates nicotine’s anti-TLR2 inhibitory effect. CFTE29o- cells ($1\times {10}^5$/mL) were left untreated or treated d-tubocurarine (d-tub, 100 $\mu$M) or α-bungarotoxin (α-bgt, 1 $\mu$M) for 16 h. Nicotine was added as indicated at 10 $\mu$M for 1 hour. Following this, some samples were stimulated with Pam₃ (1 $\mu$g/mL, 24 hours) and levels of IL-8 in supernatants were measured by ELISA (* and ${}^{\#}P\le .05$, * versus control, $\#$ or ns versus Pam₃). Assays were performed in triplicate $(n=3)$.
