Effects of p110 inhibitors on flagellin-induced Akt, p38, and ERK activation. Caco-2 cells were serum-starved overnight, and then given serum-containing media with 10 μM PI-103 (P) or TGX-221 (T) or an equivalent volume of DMSO vehicle (D). After 30 minutes, flagellin was added to a final concentration of 100 ng/ml, and cells were harvested after 1, 2, or 4 hours. Total cell lysates were separated by SDS-PAGE and tested for pSer473-Akt, phospho-p38, and phospho-ERK by Western blot. Blots were stripped and probed with total Akt and total p38 antibodies as loading controls. Results shown are typical of three independent experiments.
